Copying downloaded videos to play in iTunes is very simple; all you need to do is point iTunes to your multimedia folder or separately adding the downloaded video files to the iTunes library, and from there you can play them. As long as your downloaded videos are in supported file formats specified above (*.mp4, *.avi…etc), you can play them with iTunes without problem. There are other downloadable content over the internet that aren’t supported by iTunes however. You need to first convert them to a format supported by iTunes before you can play them. An example would be the *.mkv (Matroska) which is also a universal file format for storing multimedia data, which by using Perian, you can convert *.mkv to *.mov and play it with your iTunes.
